Effects of Sucralose Ingestion on Fetal and Placental Weights and Umbilical-Cord Length: Experimental Study International Journal of Morphology
Rodero,Ademir B; Batigália,Fernando; Azoubel,Reinaldo; Moura,Arianne Assis; Rodero,Lucas de Souza; Silveira Jr,Landulfo.
Sucralose is a sweetener of general use in over three thousand products (beverages, foods and medical diets), with utilization currently permitted for the general population in more than 80 countries, including for women of reproductive age. There is little information on the safe use of sucralose, and experimental studies have suggested that the administration of some sweeteners, by diverse routes, retards not only fetal and placental development but also umbilical cord size. The present study aimed to estimate the fetal and placental weights and umbilical-cord length in fetuses from rat dams, after ingestion and non-ingestion of sucralose. In the treated group (5 dams), sucralose was administered (via gavage) at the dose of 30 mg/kg/day, from the 10th to...

Isolation and characterization of mesenchymal stem cells derived from bovine Wharton's jelly and their potential for use in cloning by nuclear transfer Ciência Rural
Silva,Carolina Gonzales da; Martins,Carlos Frederico; Cardoso,Tereza Cristina; Cunha,Elisa Ribeiro da; Bessler,Heidi Christina; McManus,Concepta Margaret; Pivato,Ivo; Báo,Sônia Nair.
ABSTRACT: Wharton's jelly is a source of mesenchymal stem cells (MSCs) that had not yet been tested for bovine embryo production by nuclear transfer (NT). Thus, the objective of this study was to isolate, characterize and test MSCs derived from Wharton's jelly for embryo and pregnancy production by NT in cattle. The umbilical cord was collected during calving and cells derived from Wharton's jelly (WJCs) were isolated by explant and cultured in Dulbecco's Modified Eagle Medium. Skin Fibroblasts (FB) were isolated after 6 months of life. Morphological analysis was performed by bright field and scanning electron microscopy (SEM) during cell culture. Phenotypic and genotypic characterization by flow cytometry, immunocytochemistry, RT-PCR and differentiation...

PCR-based identification of Neospora caninum in the umbilical cord of a newborn calf in Brazil Ciência Rural
Azevedo da Cunha Filho,Nilton; Oliveira,Plínio Aguiar de; Oliveira,Fernando Caetano de; Pappen,Felipe Geraldo; Aguiar,Cintia Lidiane Guidotti; Santos Junior,Alceu Gonçalves dos; Costa-da-Silva,André Luis; Leite,Fabio Pereira Leivas; Farias,Nara Amélia da Rosa.
ABSTRACT: This study was conducted at a beef cattle breeding farm in the far southern region of Brazil. The birth of a calf with unilateral corneal opacity was immediately reported to the Laboratory of Parasitology, in the Universidade Federal de Pelotas, Rio Grande do Sul, Brazil; blood was collected from the cow and calf before colostrum intake. The umbilical cords from this calf and from six other healthy animals were collected. Serological examination, utilizing an indirect fluorescent antibody test, was done using a cut-off point of 1:100. Polymerase chain reaction (PCR) was also performed using the umbilical cord samples. Serological tests showed that the calf was positive for Neospora caninum at birth, with a titer of 1:1600; a titer of 1:3200 was...

Plasma amino acids in pregnancy, placental intervillous space and preterm newborn infants BJMBR
Camelo Jr.,J.S.; Martinez,F.E.; Gonçalves,A.L.; Monteiro,J.P.; Jorge,S.M..
Plasma amino acid levels have never been studied in the placental intervillous space of preterm gestations. Our objective was to determine the possible relationship between plasma amino acids of maternal venous blood (M), of the placental intervillous space (PIVS) and of the umbilical vein (UV) of preterm newborn infants. Plasma amino acid levels were analyzed by ion-exchange chromatography in M from 14 parturients and in the PIVS and UV of their preterm newborn infants. Mean gestational age was 34 ± 2 weeks, weight = 1827 ± 510 g, and all newborns were considered adequate for gestational age. The mean Apgar score was 8 and 9 at the first and fifth minutes. Plasma amino acid values were significantly lower in M than in PIVS (166%), except for aminobutyric...

Study of comparative proteome between normal and inverted karyotypes of human mesenchymal stem cells Biological Sciences
de Souza Santos, John Lenon; Câmara, Alice Barros; Meira, Isabella Tanus Job e; Oliveira, Jonas Ivan Nobre.
Multipotent mesenchymal stem cells have been expanded in vitro for cellular therapy in numerous clinical settings without standardized culture conditions or quality-control schemes. The in vitro expansion is necessary to obtain sufficient cells for clinical applications. However, the expansion may induce genetic and functional abnormalities which may affect the safety and functionality of MSC, especially the chromosomal stability. This study aimed to investigate the protein profile of umbilical cord-derived MSC with normal and inverted karyotypes after expansion in the laboratory. Mass spectrometry analysis was performed and the Bradford method, Scaffold software, String and Cytoscape databases were employed to measure and characterize the...

The TLR7 agonist Imiquimod promote the immunogenicity of msenchymal stem cells Biol. Res.
Zhang,Li; Liu,Dan; Pu,Dan; Wang,Yanwen; Li,Li; He,Yanqi; Li,Yalun; Li,Lei; Li,Weimin.
BACKGROUND: Mesenchymal stem cells (MSCs) are considered the best candidate in stem cells therapy due to their multipotent differentiation ability, low expression of co-stimulatory molecules (CD80, CD86, CD34 and HLA-II) and immunosuppression effects on in vivo immune responses. MSCs were now widely used in clinical trials but received no encourage results. The major problem was the fate of engrafted MSCs in vivo could not be defined. Some studies indicated that MSCs could induce immune response and result in the damage and rejection of MSCs. As toll like receptors (TLRs) are important in inducing of immune responses, in this study we study the role of TLR7 in mediating the immune status of MSCs isolated from umbilical cord. RESULTS: Our results indicated...
